Calcium-activated chloride channels (CaCCs) are a heterogeneous group of ligand-gated ion channels that mediate the flow of chloride ions across cell membranes in response to increases in intracellular calcium. The main molecular components identified are members of the anoctamin (TMEM16) family, particularly TMEM16A (also known as ANO1). Activation occurs when cytosolic Ca²⁺ binds to specific sites on the channel protein, triggering conformational changes that open the channel for Cl⁻ efflux or influx. CaCCs are widely expressed in both excitable and non-excitable cells and play major roles in epithelial secretion, sensory transduction, regulation of neuronal/cardiac excitability, and vascular tone control. Dysfunction or altered expression has been implicated in diseases affecting fluid secretion, hypertension, asthma, gastrointestinal motility disorders and some cancers.
