Overview

Calcium-dependent secretion activator 1 (CADPS or CAPS1) is a neural and endocrine-specific cytosolic and peripheral membrane protein required for Ca2+-regulated exocytosis of secretory vesicles, including large dense-core vesicles containing neurotransmitters and neuropeptides. It participates in the priming and fusion of these vesicles with the plasma membrane, functioning after ATP-dependent priming and involving phosphatidylinositol 4,5-bisphosphate synthesis. CADPS1 is part of the CAPS protein family, which also includes CADPS2, and plays a key role in central neurotransmitter systems and neuropeptide regulation, notably catecholamine and dopamine release. Dysregulation or mutation in CADPS family proteins can affect neuronal function and has been implicated in neurodevelopmental disorders and neurodegenerative disease.

Mechanism of action

Drugs theoretically acting on CADPS would modulate CA2+-dependent exocytosis, possibly altering neurotransmitter or peptide release. Not a direct drug target in current pharmacology.

03

Biological functions

Exocytosis of secretory vesiclesCa2+-regulated neurotransmitter and neuropeptide releaseCatecholamine loading into dense-core vesicles (DCVs)Regulated secretion of dense-core vesiclesActivity-dependent priming of exocytosisBiogenesis or maintenance of mature secretory vesicles
04

Disease associations

Neurodegenerative disease (e.g., idiopathic Parkinson’s disease, via dopamine pathway involvement)Neurodevelopmental disorders (e.g., autism spectrum disorder via CADPS2)Jejunal SomatostatinomaProstate cancerImpaired development and function of hippocampal GABAergic systems (shown in CAPS2)
05

Safety considerations

Targeting CADPS/CAPS1 could disrupt regulated neurotransmitter and peptide release, potentially leading to broad neurological or endocrine side effects.High risk of off-target effects given its ubiquitous role in regulated exocytosis in many cell types.

Interacting drugs

No approved drugs directly targeting CADPS1; none cited in clinical drug databases as direct interactors.

Biomarkers

CADPS1 itself is not a currently established clinical biomarker for patient selection or therapy monitoring.CADPS2 overexpression/variation has been associated with autism and Parkinson’s disease; CADPS1/CAPS1 is researched, not widely used clinically.
