Overview

Calcium homeostasis modulator protein 4 (CALHM4) is a member of the CALHM family of large-pore, non-selective ion channels found in human tissues, with six closely related family members identified in humans[1][2][3][5]. CALHM4 assembles as decameric or undecameric oligomers embedded in the plasma membrane, forming wide, cylindrical pores that are permeable to monoatomic cations and possibly to ATP, though its exact activation mechanism remains unknown[2][3][4]. CALHM4 is highly expressed in the human placenta, especially in trophoblasts, and may have a specialized role in mediating nutrient or ion transport at the maternal-fetal interface, but does not display pronounced ion conductance under standard conditions and its physiological gating and regulation mechanisms are not fully characterized[2][3][4]. Unlike CALHM1, which is activated by depolarization and extracellular calcium removal, CALHM4 appears to have distinct regulatory features, and its precise biological roles and disease associations are still being elucidated[3][4].
