Calcium modulating ligand (CAMLG) is an integral membrane signaling protein required for the stability of GET1 and essential for proper calcium signaling, especially in T and B lymphocytes. It acts downstream of the T cell receptor (TCR) and upstream of calcineurin by binding to cyclophilin B and causing calcium influx. This protein is implicated in immune cell activation and survival, and it has documented roles in both normal immune function and disease processes where calcium signaling is disrupted (e.g., certain cancers, congenital disorders of glycosylation). CAMLG is recognized as a ligand for the TNF receptor family member TACI and shares some mechanistic similarity with immunosuppressive drug actions (cyclosporin A).
Drugs (e.g., cyclosporin A) binding cyclophilins can modulate this signaling pathway by inhibiting the downstream activation of calcineurin (key T-cell activation step)
