Orai2 is a protein subunit of the calcium release-activated calcium (CRAC) channel, essential for store-operated calcium entry (SOCE) into cells. It is activated by STIM proteins upon depletion of Ca²⁺ stores in the endoplasmic reticulum and enables Ca²⁺ influx across the plasma membrane, playing a critical role in immune cell function, growth factor responses, neurotransmitter signaling, and cancer progression. CRAC channel activity is regulated by fast Ca²⁺-dependent inactivation (FCDI) to prevent excessive Ca²⁺ influx. Alterations or dysregulation of *ORAI2* are linked with immunodeficiency syndromes and cancer.
