The calcium-sensing receptor (CaSR) is a Class C G-protein coupled receptor (GPCR) that plays a central role in maintaining extracellular calcium ion (Ca2+) homeostasis. It senses small fluctuations in extracellular Ca2+ concentrations and modulates physiological responses accordingly. Dysfunction or mutations can lead to various calcium-related disorders.
Allosteric modulators affect the CaSR by stabilizing either the active or inactive receptor conformation, thus modulating its sensitivity to calcium. Agonist binding leads to compaction/dimerization via TM6 helix rearrangements.
