Overview

Calcium voltage-gated channel auxiliary subunit gamma-2 (CACNG2), also known as stargazin or TARP gamma-2, is a type I transmembrane protein primarily expressed in the brain, where it acts as an auxiliary subunit of voltage-gated calcium channels and as a regulatory protein for AMPA-type glutamate receptors[1][2][3][7]. CACNG2 facilitates trafficking and synaptic targeting of AMPA receptors and modulates their gating kinetics, playing a vital role in synaptic plasticity, learning, memory, and neuronal excitability[2][4][7]. Mutations or dysregulation of CACNG2 are associated with seizure disorders, intellectual disability, autism, and altered pain perception in both humans and animal models[1][3]. No specific drugs are currently known to target CACNG2 directly, but its modulation impacts AMPA receptor function and calcium channel activity, making it an emerging target for neurological and psychiatric disorders.
