Calcium voltage-gated channel auxiliary subunit gamma 3 (CACNG3) is a type I transmembrane protein that serves as an auxiliary (gamma) subunit for voltage-gated calcium channels and also functions as a regulatory protein for AMPA-type glutamate receptors, known as a transmembrane AMPAR regulatory protein (TARP). CACNG3 plays a crucial role in stabilizing the inactive state of voltage-gated calcium channels and modulates AMPA receptor trafficking to the membrane and their gating properties, thereby regulating synaptic transmission and neuronal excitability. The gene is primarily expressed in the brain and is associated with epilepsy syndromes (notably juvenile absence epilepsy), as well as implicated in neurodevelopmental disorders and potentially glioma prognosis[1][2][3][4][6]. No direct drug interactions are currently annotated, but its central role in synaptic function and disease association make it a candidate target for future therapeutics.
