Calcium voltage-gated channel auxiliary subunit gamma-6 (CACNG6) is an integral membrane protein that functions as an auxiliary subunit of voltage-gated calcium channels, particularly L-type channels. It contributes to modulation of channel activity by stabilizing the channel in an inactive (closed) state and may play additional roles in neuronal signaling and AMPA receptor regulation. CACNG6 is part of the PMP-22/EMP/MP20 protein family and has implications in several diseases, including arrhythmogenic right ventricular cardiomyopathy, depressive disorder, and aspirin-intolerant asthma. Although it is a bona fide molecular target as an ion channel auxiliary subunit, there are currently no drugs known to specifically target CACNG6[1][3][8].
