CALCRL and TFPI antisense RNA 1 (CALCRL-AS1) is a long non-coding RNA gene located at chromosome 2q32.1. It is classified as an lncRNA, and based on its name, is transcribed antisense to both the CALCRL (Calcitonin receptor-like receptor) and TFPI (Tissue factor pathway inhibitor) loci. Antisense lncRNAs typically regulate the expression of neighboring or overlapping genes by a variety of mechanisms, including transcriptional interference, epigenetic modification, and RNA-RNA interactions[2]. CALCRL-AS1 has been associated in genetic studies with phenotypes such as lymphatic malformation, suggesting some regulatory or disease relevance, but its precise biological function and role in pathology are not well defined[3][9]. Unlike protein-coding genes, CALCRL-AS1 is not a receptor, enzyme, or transporter, and is not known to be a direct target of any approved drugs. Note: - CALCRL-AS1 is not to be confused with *CALCRL* (Calcitonin receptor-like receptor), which is a well-characterized G protein-coupled receptor and a known drug target in migraine (notably antagonized by CGRP-targeting drugs)[1][6][7][10]. - This entry is specific to the lncRNA CALCRL-AS1, not the protein-coding CALCRL. - There is no evidence that CALCRL-AS1 is itself a therapeutic target, even though the protein-coding gene CALCRL is. - For current research or clinical purposes, CALCRL-AS1 should be considered a gene of interest primarily for its regulatory potential in gene expression rather than as a direct target for drugs or therapies.
No mechanism of action for drugs, as no drugs are reported to directly target CALCRL-AS1. Theoretical methods could include antisense oligonucleotides or RNA interference approaches, generically applied to non-coding RNAs.
