Calcyclin-binding protein (CACYBP) is a modular adaptor or scaffold protein implicated in multiple cellular processes, including protein dephosphorylation, ubiquitination, cytoskeletal organization, and signaling pathways regulating cell proliferation and differentiation[1][2][4][5]. CACYBP binds to calcium-binding proteins such as S100A6, components of the ubiquitin ligase complex (including Siah-1), and cytoskeletal proteins, participating in calcium-dependent ubiquitination and subsequent proteasomal degradation of target proteins[1][4][5]. It is highly expressed in tumor cells and neurons, with dysregulation associated with certain cancers and neurodegenerative diseases, such as Alzheimer’s disease[1]. While it is not a classic receptor, enzyme, or transporter, CACYBP plays a key role in cell homeostasis and signaling; its association with tumorigenesis and neurodegenerative pathologies highlights its potential as a therapeutic and biomarker target[1][2][4].
