Calcyphosine is a calcium-binding protein with four EF-hand domains, originally identified as thyroid protein p24 and also found in epithelial cells and some central nervous system tissues. It undergoes calcium-dependent conformational changes and is synthesized/phosphorylated in thyroid follicular cells upon thyrotropin and cAMP stimulation. Calcyphosine may participate in the regulation of ion transport, possibly crosslinking calcium and cyclic AMP signaling pathways. Its overall structure is similar to calmodulin but shows unique features among the EF-hand protein family. While not a receptor or classic drug target, calcyphosine has been studied for its possible roles in cell proliferation, microtubule dynamics, and as a modulator of kinase signaling, and is upregulated in some cancers (notably, glioma and ependymoma). It has no known direct pharmacological modulators or drugs.
