Overview

Calicin (CCIN) is a basic, testis-specific cytoskeletal protein and a major structural component of the perinuclear theca (PT), specifically the postacrosomal "calyx" region of the mammalian sperm head[2][3]. It is characterized by kelch repeats and a BTB/POZ domain and shows strong homology to other actin-associated proteins, suggesting a role in actin network organization and cytoskeletal stability[3][2]. Calicin stabilizes the structural integrity and precise morphology of the sperm head during spermatogenesis[1][2]. Its expression is restricted to testicular germ cells, and it is essential for male fertility; mutations or dysregulation can cause acrosome detachment, sperm head deformities, and male infertility such as globozoospermia[1][2][3]. The protein is not considered a classical therapeutic target such as a receptor or enzyme, and there are no known drugs targeting CCIN. However, defective CCIN protein or gene mutations serve as biomarkers for specific types of male infertility.
