Overview

Calmegin is a testis-specific calcium-binding molecular chaperone protein localized to the endoplasmic reticulum of meiotic and post-meiotic germ cells. Its principal function is to assist in folding and assembly of key sperm membrane proteins, notably those required for sperm migration and sperm-zona pellucida binding during fertilization. Knockout mouse models lacking calmegin display male infertility due to defective processing of sperm membrane proteins rather than abnormalities in germ cell morphology. Calmegin expression is regulated throughout spermatogenesis, particularly during the stages of high sperm protein synthesis. There are no approved therapies targeting calmegin, and its role is restricted to male reproductive biology as a chaperone protein rather than as an enzyme, receptor, or transporter.
