Overview

Calmodulin-like protein 5 (CALML5) is a calcium-binding protein of the calmodulin family, primarily expressed in the epidermis and specifically in differentiating keratinocytes. CALML5 undergoes conformational changes upon binding calcium and plays an important role in terminal differentiation of keratinocytes, contributing to the formation and maintenance of the epidermal barrier. It is involved in regulating the expression of late differentiation genes, a process controlled by the ZNF750 transcription factor and stabilized by the long non-coding RNA TINCR. CALML5 interacts with proteins such as transglutaminase 3 and stratifin (SFN/14-3-3σ), modulating cell adhesion and protein complexes involved in skin differentiation and barrier function. Downregulation or loss of CALML5 impairs skin differentiation; it is notably reduced in squamous cell carcinoma. There are currently no known drugs that target CALML5, nor established safety concerns or drug mechanisms related to this molecule.
