Overview

Calmodulin-regulated spectrin-associated protein 2 (CAMSAP2) is a microtubule minus-end binding protein crucial for the organization and stabilization of non-centrosomal microtubule arrays. CAMSAP2 regulates cytoskeletal architecture by binding directly to microtubule minus ends through its CKK domain, autonomously stabilizing the lattice and protecting ends from depolymerization. The protein also orchestrates microtubule nucleation—independent of the classical γ-tubulin ring complex—by promoting condensate formation with tubulin and acting as a nucleation center, essential for aster-like and meshwork microtubule arrangements in cells. CAMSAP2 is involved in processes such as cell migration, polarization, Golgi organization, insulin secretion in pancreatic β-cells, and blood-testis barrier dynamics during spermatogenesis[1][2][3][4][5]. It is a structural and functional regulator of cytoskeletal microtubules, but is not currently recognized as a direct therapeutic target or drugged protein.
