Overview

Calneuron-1 is a calcium-binding protein highly similar to members of the calmodulin family and characterized by the presence of EF-hand motifs, some of which are non-functional[1][2][3]. It is a neuron-specific protein that serves as a calcium sensor and acts as a membrane-anchored regulator, particularly at the trans-Golgi network and Golgi membranes, where it controls vesicle trafficking from the Golgi to the plasma membrane by regulating phosphatidylinositol 4-kinase IIIβ[1][3]. Calneuron-1 is implicated in both neuronal signaling and cancer biology, influenced by various microRNAs, and shows specific subcellular localization and functional roles distinct from other calcium sensor proteins. Genetic variation in CALN1 has been associated with susceptibility to neuropsychiatric features such as impaired sensorimotor gating[1]. There are currently no known drugs that directly target calneuron-1, and its role as a therapeutic target is not established.
