Calpain 1 is a calcium-activated, non-lysosomal cysteine protease that plays a key role in various cellular processes, including cytoskeletal remodeling and signal transduction. It functions as part of a heterodimeric complex composed of a large catalytic subunit (encoded by CAPN1) and a common small regulatory subunit (CAPNS1/CAPN4). Activation requires micromolar concentrations of intracellular Ca²⁺ ions. Dysregulation or aberrant activation has been implicated in several diseases, including Alzheimer's disease.
