Calpain-6 (CAPN6) is an atypical, non-proteolytic member of the calpain family of calcium-dependent cysteine proteases, but unlike most calpains, it lacks proteolytic activity due to active site mutations[1][2][3][5][6]. CAPN6 is highly expressed in embryonic muscle and placenta and acts as a multifunctional regulator of cytoskeletal dynamics, microtubule stabilization, and cell migration, where it can influence actin organization and cell differentiation. In muscle, it suppresses myogenic differentiation and regeneration. CAPN6 is also implicated in the inhibition of apoptosis and promotion of angiogenesis in tumorigenesis, suggesting potential relevance to cancer. Its unique absence of protease activity distinguishes it within the calpain family, and it has been proposed as a possible therapeutic target in certain muscle diseases and cancers[1][2][3][5][6].
