Overview

Calpain-8 (CAPN8) is a calcium-regulated, non-lysosomal thiol-protease belonging to the calpain family of cysteine proteases. It is primarily involved in proteolysis and membrane trafficking, especially within gastric pit cells (surface mucus cells) in the stomach. Biochemically, calpain-8 requires calcium as a cofactor for its enzymatic activity and participates in the physiological turnover and regulation of cellular substrates[1][3][4]. This protein is encoded by the CAPN8 gene on human chromosome 1[1][3]. While recognized as a biologically active enzyme, calpain-8's direct involvement in specific human diseases and its role as a drug target are not substantiated by current literature[1][3][4].
