Calponin-2 is a 33.7 kDa actin-binding regulatory protein broadly expressed in smooth muscle, fibroblasts, endothelial cells, epithelial cells, immune cells, and various cancer cells. It stabilizes actin filaments, inhibits actin-activated myosin ATPase, and modulates cell mechanics, migration, proliferation, and energy metabolism. In disease, CNN2 is implicated in cancer cell invasion, modulation of inflammation and fibrotic responses, and is upregulated in arthritic and fibrotic tissues. Though not yet targeted by approved drugs, its biological significance makes it an attractive candidate for therapeutic intervention.
