Overview

Calsyntenin-1 is a type-1 neuronal transmembrane protein of the cadherin superfamily encoded by the CLSTN1 gene in humans[2][3]. Also known as alcadein-alpha, it functions as a postsynaptic adhesion molecule that binds presynaptic neurexins, thereby mediating excitatory and inhibitory synapse formation[2]. It plays a key role in axonal anterograde transport by acting as a cargo adaptor linking specific vesicles to the kinesin-1 motor complex[1][2]. In addition, it forms complexes with amyloid precursor protein (APP) and APBA2, modulating APP trafficking and amyloid-beta processing, processes implicated in the pathogenesis of Alzheimer's disease[2][3]. Calsyntenin-1 binds calcium ions and is involved in protein-protein interactions, including with APP and APBA2, but is not currently considered a therapeutic target such as a receptor, enzyme, or transporter[2][3]. No drugs are known to interact directly with Calsyntenin-1, and it is not used as a biomarker or associated with specific safety concerns.
