Overview

CaM kinase-like vesicle-associated protein (CAMKV) is a membrane-associated calmodulin-binding pseudokinase of the CaM kinase family, lacking intrinsic kinase activity but binding calmodulin in a calcium-dependent manner[1][2][3]. It is expressed in neuronal tissue, where it regulates dendritic spine maintenance and synaptic plasticity by modulating RhoA signaling through direct interaction with the Rho guanine nucleotide exchange factor Lfc[3]. CAMKV is also transcriptionally regulated by MYCN and highly expressed on the cell surface of MYCN-amplified neuroblastoma, making it a candidate immunotherapeutic target for these cancers[2]. Outside the CNS, CAMKV expression is limited, but it has been proposed as a biomarker in paraneoplastic neurological disorders[6]. There are currently no approved drugs targeting CAMKV, but antibody-based approaches are under investigation for tumor selectivity[2].

Mechanism of action

No currently approved therapeutic drugs or biologics directly act on CAMKV in clinical use. Proposed as an immunotherapy target (antibody or antibody-drug conjugate) in MYCN-amplified neuroblastoma due to cell surface expression in tumors and low expression outside central nervous system
