cAMP-dependent protein kinase inhibitor gamma (PKIG) is a regulatory protein encoded by the PKIG gene, classified as a member of the protein kinase inhibitor family. It acts as a potent competitive inhibitor of cAMP-dependent protein kinase (PKA) by binding the catalytic subunit after cAMP-induced dissociation of regulatory chains, thereby modulating kinase activity. PKIG is widely expressed, especially in heart, skeletal muscle, and testis, as well as in endothelial cells across multiple vascular beds, where it influences endothelial barrier function and cAMP-mediated signaling. PKIG also plays a key regulatory role in mesenchymal cell differentiation, with downregulation required for osteogenic activation. While no approved drugs target PKIG directly, its modulation is relevant to understanding PKA-dependent signal transduction, lineage specification, and potentially immune function.
