Overview

cAMP-regulated phosphoprotein 19 (ARPP19) is an intrinsically disordered phosphoprotein that functions as a specific inhibitor of protein phosphatase 2A (PP2A) during mitosis[1][2][3]. ARPP19 is phosphorylated by kinases such as MASTL, and the phosphorylated form binds to the B55 regulatory subunit of PP2A, leading to inhibition of the phosphatase and regulation of G2/M cell cycle transition[1]. The ARPP19/PP2A axis is critical for mitotic progression and cell cycle control. Structurally, ARPP19 lacks a stable 3D fold but contains transient helices and engages in multiple conformational states[2]. Dysfunction or dysregulation of ARPP19 has been associated with disease states including cancer and certain brain malformation syndromes[2][3].
