Overview

cAMP responsive element binding protein 5 (CREB5) is a transcription factor belonging to the CREB (cAMP response element-binding) protein family, characterized by a basic leucine zipper (bZIP) DNA-binding domain.[2][3] It binds to the cAMP response element (CRE) as a homodimer or heterodimer (notably with c-Jun or CRE-BP1)[2], acting as a CRE-dependent trans-activator involved in the regulation of target gene expression. CREB5 plays a critical role in synovial joint formation and articular cartilage development by directing gene programs that guide tissue differentiation[1]. In cancer biology, CREB5 reprograms FOXA1 nuclear interactions and has been implicated in promoting resistance to androgen receptor-targeting therapies in prostate cancer[6]. CREB5 also contributes to cell differentiation (e.g., white fat cell differentiation) and is implicated in certain inflammatory diseases, including inflammatory bowel disease[2][5][6]. No approved drugs directly target CREB5 as of current knowledge, and its modulation as a therapeutic approach remains at the investigational stage.
