Overview

The lysosome is a membrane-bound organelle in eukaryotic cells responsible for degrading and recycling macromolecules. In cancer, lysosomes support increased metabolic demands, cell survival under stress, tumor progression, and therapy resistance. It is the target for cancer therapeutics such as antibody-drug conjugates (ADCs), that release their cytotoxic payload within the lysosome. Targeting the lysosome enables selective intracellular drug release using acid-sensitive or enzyme-cleavable linkers. However, lysosomes have a dual role in tumor biology; they promote autophagic clearance but also support metabolic adaptation and enable invasion/metastasis.
