Cancer susceptibility 10 (CASC10), also known as LINC00340, is a long non-coding RNA (lncRNA) located on chromosome 10q26 that functions as a critical oncogenic driver in several human cancers (NCBI Gene ID: 100506658). It operates primarily through a competitive endogenous RNA (ceRNA) mechanism, where it acts as a 'sponge' for tumor-suppressive microRNAs such as miR-370, miR-543, and miR-129-5p, thereby derepressing oncogenic target genes (PubMed: 31480385, PubMed: 30899214). Elevated expression of CASC10 is strongly associated with advanced clinical stage, lymph node metastasis, and poor overall survival in patients with gastric, colorectal, and hepatocellular carcinomas (PubMed: 32440145). In addition to its role in cell proliferation and apoptosis resistance, CASC10 promotes the epithelial-mesenchymal transition (EMT), facilitating tumor invasion and systemic spread. While no small molecule or biologic drugs are currently approved for CASC10, it is a high-priority target for RNA-targeted therapeutic modalities, such as antisense oligonucleotides (ASOs) and siRNA-based knockdown strategies, which aim to neutralize its oncogenic activity. The development of such therapies faces typical challenges for RNA therapeutics, including the need for stable delivery systems and the mitigation of off-target effects within the complex landscape of non-coding RNA networks.
