Overview

Cancer susceptibility candidate 10 (CASC10) is a long non-coding RNA (lncRNA) gene located on chromosome 9p21.3, a genomic region heavily implicated in susceptibility to various cancers and cardiovascular diseases (HGNC, 2024; NCBI Gene, 2024). It resides within a cluster of critical tumor suppressors, including CDKN2A and CDKN2B, and is often co-regulated with the antisense RNA ANRIL (PubMed, PMID: 29156761). CASC10 is thought to play a role in the epigenetic and transcriptional regulation of these neighboring genes, thereby influencing cell cycle progression and cellular senescence (PubMed, PMID: 31434980). Dysregulation of CASC10 has been linked to increased risk and progression of conditions such as glioma, melanoma, and coronary artery disease (PubMed, PMID: 30262823). While not yet a target for approved small-molecule drugs, it represents a potential target for RNA-based therapeutic interventions like antisense oligonucleotides (ASOs) or siRNA (PubMed, PMID: 28445534). Understanding its precise molecular interactions is crucial for developing precision medicine strategies for individuals carrying risk alleles at the 9p21.3 locus.
