Overview

NY-ESO-1, also known as Cancer-testis antigen 1B (CTAG1B), is a member of the cancer-testis antigen (CTA) family. It is a highly immunogenic protein that has become a prominent target for cancer immunotherapy due to its unique expression profile and ability to elicit strong immune responses. Expression is restricted to testis in normal tissues, but is aberrantly re-expressed across various malignancies, making it an attractive target. The precise physiological role of NY‑ESO‑1 remains incompletely understood, but it may be involved in gametogenesis and cell cycle progression. Immunotherapeutic approaches target NY-ESO-1 via peptide vaccines, adoptive T-cell therapies, and checkpoint inhibitor combinations.
