Overview

Cancer-testis antigen 1B (NY-ESO-1) is a highly immunogenic tumor-associated antigen encoded by the CTAG1B gene on chromosome Xq28 and is composed of 180 amino acids (18 kDa)[2][3]. NY-ESO-1 shows restricted expression in normal adult tissues (mainly germ cells of the testis and placenta) but is aberrantly expressed in a wide range of cancers, making it an attractive target for cancer immunotherapy, especially in melanoma, synovial sarcoma, neuroblastoma, and epithelial ovarian cancers, among others[1][2][3][6]. Its biological function remains incompletely understood but may relate to cell cycle regulation, cell proliferation, and germ cell biology, with evidence of nuclear versus cytoplasmic localization patterns in different cell types[2][5]. NY-ESO-1–based immunotherapies—including vaccines and adoptive T cell treatments targeting its epitopes—have shown promise in clinical trials, largely owing to the protein's ability to induce robust humoral and cellular immune responses without significant risk of targeting most healthy tissues[1][3][7].

Mechanism of action

Immune recognition: therapies induce or transfer NY-ESO-1–specific T cells to target and eliminate NY-ESO-1–expressing tumor cells Cancer vaccines aim to elicit specific anti-NY-ESO-1 T cell and antibody responses for tumor eradication

03

Biological functions

Cell cycle regulation (putative, based on domain composition and expression pattern)Cell proliferation (suggested by expression in stem and tumor cells)Possible role in germ cell differentiation or self-renewalElicits strong humoral and cellular immune responses
04

Disease associations

Cancer (including melanoma, synovial sarcoma, neuroblastoma, liposarcoma, ovarian cancer, lung cancer, bladder cancer, among others)Tumor biomarkerOther (limited: marker in testicular germ cell tumors)
05

Safety considerations

Off-tumor expression in male germ cells and placental tissue (risk of infertility in males or pregnancy-associated effects; minimal in somatic tissues)Heterogeneous antigen expression in tumors may lead to immune evasion or incomplete therapeutic responsePotential for autoimmune toxicity is limited due to restricted normal tissue expression but must be monitored in clinical settings
