Cancer-testis antigen 1B (NY-ESO-1) is a highly immunogenic tumor-associated antigen encoded by the CTAG1B gene on chromosome Xq28 and is composed of 180 amino acids (18 kDa)[2][3]. NY-ESO-1 shows restricted expression in normal adult tissues (mainly germ cells of the testis and placenta) but is aberrantly expressed in a wide range of cancers, making it an attractive target for cancer immunotherapy, especially in melanoma, synovial sarcoma, neuroblastoma, and epithelial ovarian cancers, among others[1][2][3][6]. Its biological function remains incompletely understood but may relate to cell cycle regulation, cell proliferation, and germ cell biology, with evidence of nuclear versus cytoplasmic localization patterns in different cell types[2][5]. NY-ESO-1–based immunotherapies—including vaccines and adoptive T cell treatments targeting its epitopes—have shown promise in clinical trials, largely owing to the protein's ability to induce robust humoral and cellular immune responses without significant risk of targeting most healthy tissues[1][3][7].
