Cancer-testis antigen 1B (NY-ESO-1) epitope 91-110 (NY-ESO-1 (91-110))

NY-ESO-1 (Cancer-testis antigen 1B) is a highly immunogenic protein expressed in various malignancies but restricted to germ cells in healthy adults, making it an ideal target for immunotherapy (UniProt P12023). The 91-110 epitope is a 20-amino acid peptide (YLAMPFATPMEAELARRSLA) that serves as a promiscuous immunodominant region, capable of inducing both CD4+ helper and CD8+ cytotoxic T-cell responses across multiple HLA alleles, including HLA-A*0201 and HLA-A*2402 (Eikawa et al., 2013). It has been extensively studied in cancer vaccine clinical trials, often formulated with adjuvants like ISCOMATRIX to enhance its immunogenicity (Nicholaou et al., 2004). Targeting this epitope aims to stimulate a broad anti-tumor immune response to control or eradicate NY-ESO-1-positive tumors. Clinical studies have demonstrated that vaccination with this peptide can lead to stable disease and the induction of robust cellular and humoral immunity in patients with advanced cancers (Jäger et al., 2000). Because of its ability to activate multiple arms of the immune system, it remains a key component in the development of next-generation cancer vaccines and combination therapies.
