Overview

The Candida species mitochondrial membrane is a vital organelle structure that plays a central role in the energy metabolism, stress response, and virulence of pathogenic fungi such as Candida albicans (NIH, 2023). It is composed of an outer membrane and a highly folded inner membrane, the latter containing the electron transport chain (ETC) and ATP synthase complexes responsible for oxidative phosphorylation (ASM, 2024). This membrane is a key therapeutic target because its disruption—either through direct pore formation, inhibition of respiratory complexes (e.g., Complex III or IV), or blockade of essential transporters—leads to the collapse of the mitochondrial membrane potential (MMP) and a lethal surge in reactive oxygen species (ROS) (MDPI, 2020). Several novel antifungal candidates, such as T-2307 and ilicicolin H, specifically target mitochondrial membrane components, often exploiting fungal-specific structural variations to achieve selectivity over human mitochondria (NIH, 2025). Furthermore, mitochondrial membrane function is intricately linked to azole resistance, making it a strategic target for combination therapies aimed at restoring drug sensitivity in resistant Candida strains (MIT, 2016).
