Canine malignant melanoma (CMM) cells are highly aggressive neoplastic cells derived from melanocytes, most commonly affecting the oral cavity, nail beds, and skin of dogs [5, 10]. These cells are characterized by rapid proliferation, a high propensity for metastasis, and resistance to standard radiation and chemotherapy, making them a primary challenge in veterinary oncology [2, 3]. Although 'Canine malignant melanoma cells' refers to a cell population rather than a single molecular target, they serve as the functional target for various therapeutic interventions because of their expression of specific antigens and dysregulated signaling pathways [6]. For example, the Oncept vaccine targets these cells by inducing an immune response against tyrosinase, an enzyme often overexpressed in the melanoma lineage [5]. Additionally, CMM cells frequently demonstrate overactivation of the PI3K/AKT/mTOR and MAPK/ERK pathways, which are therapeutic focal points for inhibitors like toceranib and rapamycin [1, 2, 7]. Due to their biological and genetic similarities to human mucosal melanoma, CMM cells are considered an important comparative oncology model [6, 10].
Drugs targeting these cells utilize several distinct mechanisms: the Oncept vaccine induces a xenogeneic immune response against the Tyrosinase enzyme expressed by the cells [5]; small molecule inhibitors such as Toceranib target receptor tyrosine kinases including KIT and VEGFR to inhibit growth and angiogenesis [2, 4]; rapalogs like Rapamycin and Everolimus inhibit the PI3K/AKT/mTOR signaling pathway critical for cellular survival and proliferation [1, 7]; and Selective Inhibitors of Nuclear Export (SINE) like Verdinexor trap tumor suppressor proteins within the cell nucleus to induce apoptosis [2].
