Overview

Cannabinoid receptor-interacting protein 1 (CNRIP1) is an intracellular protein that specifically binds to the C-terminal region of the cannabinoid receptor 1 (CB1R), a key G protein-coupled receptor in the endocannabinoid system[1][2][5]. CNRIP1—particularly its CRIP1a isoform—modulates both constitutive and agonist-induced CB1R signaling by acting as a negative regulator, attenuating CB1R-mediated G-protein activation and the receptor’s impact on voltage-gated calcium channels[2][3][4][8][9]. CNRIP1 is broadly expressed in the brain as well as peripheral tissues, with evidence pointing to its roles in neuronal signaling, CB1R turnover, and possibly in non-neuronal tissues[1][4][5]. It has been implicated in neurological diseases (including epilepsy and other CNS disorders) and cancers due to changes in expression or promoter methylation, although it is not itself targeted by approved drugs[1][4][6]. CNRIP1 is best understood as a regulatory protein of CB1R, indirectly influencing the pharmacodynamics of cannabinoids and related ligands, rather than as a direct therapeutic target[1][2][4].
