Overview

The cannabinoid type 1 receptor (CB1) is a G protein-coupled receptor (GPCR) encoded by the CNR1 gene. It is highly expressed throughout the CNS and also found in peripheral tissues. CB1 is activated by endogenous cannabinoids (endocannabinoids) such as anandamide and 2-arachidonoylglycerol, plant-derived phytocannabinoids like tetrahydrocannabinol (THC), and synthetic analogs of THC. It modulates neurotransmission, appetite, pain perception, and plays key roles in memory, learning, and motor control. CB1 receptors are implicated in various disease states including addiction disorders, obesity, and neuropsychiatric conditions.

Mechanism of action

Gi/o-mediated inhibition of adenylyl cyclase leading to decreased cAMP levels, activation of MAPK, inhibition of voltage-gated Ca2+ channels, activation of inwardly rectifying K+ currents, modulation of nitric oxide signaling pathways, and modulation of ion channels.
