The cap binding complex (CBC) is a highly conserved, essential heterodimeric protein complex in eukaryotes that binds to the 5' cap structure of nascent messenger RNA (mRNA). This interaction is crucial for multiple steps in gene expression, including mRNA processing, export, surveillance, and translation initiation. CBC consists of two main subunits: CBP20 (Cap Binding Protein 20; also known as NCBP2) and CBP80 (Cap Binding Protein 80; also known as NCBP1). After transcription, CBC binds cotranscriptionally to nascent pre-mRNAs at their capped 5′ end and remains associated through nuclear processing events until export into the cytoplasm. Upon entry into the cytoplasm or after pioneer round(s) of translation, CBC is replaced by eIF4E/eIF4F on most mature mRNAs.
