Overview

Capping actin protein of muscle Z-line subunit beta (CAPZB) is the β subunit of the heterodimeric F-actin capping protein, a key regulator of actin filament dynamics found in nearly all eukaryotic cells[1][4]. CAPZB binds to the barbed (rapidly growing) ends of actin filaments, blocking both their assembly and disassembly, thus stabilizing filament length and organizing the cytoskeleton in muscle and nonmuscle cells[1][3]. In muscle, CAPZB is anchored at the Z-disk, where it maintains sarcomere structure essential for muscle contraction and prevents mechanical stress-induced damage[2][3][5]. Beyond muscles, CAPZB is also required for processes like cell morphology maintenance, differentiation, neural crest migration, and auditory function by governing the architecture of branched actin networks and stereocilia[2][4][5]. Disruption or dysregulation of CAPZB is associated with developmental disorders such as craniofacial anomalies and muscle malformation, but it is not a current direct therapeutic target[1][2][3][7].
