Overview

Caprin family member 2 (CAPRIN2) is a multifunctional cytoplasmic protein acting primarily as an RNA-binding and scaffolding molecule. It plays a pivotal role in the regulation of the canonical Wnt/β-catenin signaling pathway by facilitating phosphorylation of the Wnt co-receptors LRP5/6, which stabilizes β-catenin and activates Wnt target gene transcription[1][7]. CAPRIN2 functions are highly context-dependent and include regulation of cell proliferation, differentiation (especially in erythroblasts and lens fiber cells), and involvement in tissue-specific developmental processes[1][4][6]. Structurally, CAPRIN2 carries homologous domains with its family member Caprin-1, and forms homodimers via its HR1 domain[3][7], potentially mediating formation of specialized ribonucleoprotein complexes. In the brain, CAPRIN2 regulates mRNA stability and plasticity in hypothalamic neurons, critically mediating responses to osmotic challenges and influencing vasopressin transcript abundance and modification[2][5]. Its dysregulation has been linked to developmental defects, cancer progression, multidrug resistance, and altered physiological homeostasis, but it currently lacks direct pharmacological modulation.
