The capsid scaffolding protein in Human herpesvirus 5 (HHV-5), also known as human cytomegalovirus (HCMV), is a critical viral protein required for the assembly of the viral capsid. It is encoded by the UL80.5 gene and is commonly referred to as the UL80 or capsid scaffolding protein. The primary role is to facilitate proper assembly of the icosahedral capsid shell within infected cell nuclei. It achieves this by promoting interactions with major capsid proteins, thereby guiding correct formation and stabilization of procapsids before DNA packaging occurs. During maturation, a virus-specific protease cleaves off portions of the scaffolding proteins.
