The capsid-spacer peptide 1 (CA-SP1) region is a critical segment within the Gag polyprotein of retroviruses, most notably HIV-1. The Gag polyprotein is essential for viral assembly and maturation, and its proper processing by the viral protease is required for infectivity. The CA-SP1 region specifically refers to the junction between the capsid (CA, p24) domain and spacer peptide 1 (SP1), a short sequence that plays a pivotal role in virus particle formation and maturation. It is essential for correct virion assembly and morphological transition during budding/maturation. Small molecules like bevirimat inhibit HIV replication by blocking cleavage at CA-SP1. This prevents proper core condensation and halts virus maturation—making it an attractive antiviral target.
