Overview

The capsular polysaccharides of Neisseria meningitidis are complex carbohydrate polymers that form the outermost protective layer of the bacterium. These polysaccharides are the primary determinants of the 12 different serogroups of N. meningitidis and serve as critical virulence factors by allowing the pathogen to evade host immune responses, such as complement-mediated killing (Stephens et al., 2007). Serogroups A, C, Y, W-135, and X are globally significant as they are responsible for the majority of invasive meningococcal disease cases, including life-threatening meningitis and septicemia (WHO, 2023). In pharmaceutical development, these polysaccharides are utilized as antigens in vaccines to induce protective immunity. Modern conjugate vaccines chemically link these polysaccharides to carrier proteins to enhance immunogenicity and induce long-term immunological memory (Pollard et al., 2009). Upon vaccination, the immune system produces specific antibodies that recognize these polysaccharides, leading to the destruction of the bacteria through opsonization and complement activation (Borges et al., 2019).
