Carbohydrate-binding module 62 (CBM62) is a family of non-catalytic protein domains found within various carbohydrate-active enzymes (CAZymes), primarily in bacteria such as Clostridium thermocellum and Bacteroides plebeius (CAZy Database, 2024; Cazypedia, 2021). These modules are characterized by a beta-jelly-roll fold and specifically bind to terminal D-galactose and L-arabinopyranose residues on complex polysaccharides like xyloglucan, galactomannan, and arabinogalactan (Cazypedia, 2021; Journal of Biological Chemistry, 2011). By tethering the associated catalytic enzyme to its substrate, CBM62 increases the local enzyme concentration and enhances the efficiency of polysaccharide degradation (Biochemical Journal, 2004; Cazypedia, 2023). In the context of human health, CBM62-like domains have been identified in virulence factors of pathogens such as Streptococcus oralis (AsaA) and Streptococcus pyogenes (EndoS), where they may facilitate bacterial adhesion to host tissues or the processing of host glycans (PLOS Pathogens, 2020; Journal of Biological Chemistry, 2011). Additionally, CBM62 plays a role in the human gut microbiome's ability to metabolize specific dietary fibers, such as those found in seaweed, which can influence host nutrition and intestinal health (Nature, 2010; Microbiome Atlas, 2024). While CBM62 is not currently the target of any approved drugs, its role in bacterial pathogenesis and microbiome function makes it a subject of interest for potential anti-virulence strategies and nutritional interventions (PLOS Pathogens, 2020; Nature Reviews Microbiology, 2013).
