Overview

Sugar metabolism refers to the collective biochemical processes responsible for the breakdown, synthesis, and interconversion of sugars, primarily glucose, to provide energy for cellular functions. This complex network includes pathways such as glycolysis, the citric acid cycle, gluconeogenesis, and glycogen metabolism, all of which are tightly regulated by hormones like insulin and glucagon (StatPearls, 2023). Dysregulation of these pathways is a central feature of numerous metabolic disorders, most notably diabetes mellitus, where impaired glucose uptake or excessive endogenous production leads to chronic hyperglycemia (NIH, 2022). While 'Sugar metabolism' is a broad physiological process rather than a single molecular target, many of its constituent enzymes and transporters serve as critical points for pharmacological intervention (PubChem, 2023). For example, biguanides like metformin target mitochondrial processes to reduce hepatic glucose production, while SGLT2 inhibitors target renal transporters to increase glucose excretion (PubMed, 2021). Understanding the flux of these pathways is essential for developing therapies for metabolic syndrome, cardiovascular disease, and certain types of cancer that exhibit altered glucose utilization (Wikipedia, 2023).

Mechanism of action

Modulation of glucose levels through the inhibition of hepatic gluconeogenesis, enhancement of peripheral insulin sensitivity, inhibition of renal glucose reabsorption, and delay of intestinal carbohydrate absorption (StatPearls, 2023).
