Carbohydrate sulfotransferase 3 (CHST3) is an enzyme that catalyzes the transfer of sulfate from 3'-phosphoadenosine 5'-phosphosulfate (PAPS) to the 6-position of N-acetylgalactosamine residues within chondroitin, resulting in the formation of chondroitin 6-sulfate—a major proteoglycan in cartilage and extracellular matrices[1][2][3][5][6]. This modification is essential for the development, maintenance, and biomechanical integrity of cartilage and bone, supports cellular migration and differentiation, and plays a role in immune cell maintenance in the spleen[1][2][3][5][8]. CHST3 deficiency or pathogenic variants can lead to rare skeletal dysplasias presenting with short stature, progressive joint dislocations, and bone deformities; it may also be involved in intervertebral disc degeneration and other connective tissue diseases[2][5][6][7]. No approved drugs directly target this enzyme, though its activity may serve as a potential biomarker and therapeutic target for degenerative skeletal disorders.
