Carbonic anhydrase 2 (CA2), also known as carbonic anhydrase II, is a zinc-containing metalloenzyme that catalyzes the reversible hydration of carbon dioxide to bicarbonate and a proton, playing a crucial role in physiological processes such as respiration, pH regulation, ion transport, and bone resorption.
The enzyme uses its zinc-bound hydroxide for nucleophilic attack on CO₂ to form bicarbonate; this is then exchanged for water. A subsequent proton transfer regenerates the active site hydroxide via solvent exchange—a classic ping-pong mechanism.
