Carbonic anhydrase-related protein 11 (CA11) is an evolutionarily conserved, secreted protein encoded by the CA11 gene in humans and classified within the carbonic anhydrase family; unlike the enzymatically active members, CA11 is "acatalytic," lacking the necessary residues for zinc binding and catalytic hydration of carbon dioxide[1][2][3]. It is predominantly expressed in the brain—especially in neuronal cell bodies, neurites, and astrocytes—and evidence suggests roles in neuronal development, differentiation, and synaptic regulation, including functioning as a neurexin ligand; CA11 is implicated in neurodevelopment, synaptic activity modulation, and tumor biology, though it does not appear to be a direct therapeutic target or established biomarker in clinical use[2][3]. Reduced expression of CA11 is linked to higher glioma grade and poor patient outcomes, while altered distribution is observed in neurodegenerative models, and its biological actions appear to be mediated primarily by protein–protein interactions rather than enzyme activity[2].
