Overview

Carbonic anhydrase XII (CAXII) is a zinc metalloenzyme that catalyzes the reversible hydration of carbon dioxide to bicarbonate and protons. As a type I membrane protein with its catalytic domain exposed extracellularly, CAXII plays a crucial role in pH regulation and CO₂ homeostasis. It is involved in various biological functions, including fluid secretion and maintenance of physiological pH across tissues. CAXII is overexpressed in some cancers, particularly under hypoxic conditions, supporting tumor growth and metastasis. Small-molecule inhibitors, such as sulfonamides, and monoclonal antibodies, like 6A10, can inhibit CAXII activity, offering potential therapeutic strategies for cancer treatment and other diseases associated with its dysregulation.
