Overview

Carboxy-terminal domain RNA polymerase II polypeptide A small phosphatase 2 (CTDSP2) is a nuclear phosphatase enzyme that specifically dephosphorylates the Ser-5 residue in the C-terminal domain (CTD) repeats of RNA polymerase II, thereby regulating RNA polymerase II activity and gene transcription. CTDSP2 plays a multifunctional role, influencing cell proliferation, matrix remodeling, and transcriptional regulation, including both direct impact on cell cycle checkpoints through Ras signaling and p21 upregulation, as well as modulation of TGF-β/SMAD signaling and extracellular matrix production. It is implicated in oncogenic processes, possibly acting as a tumor suppressor, and interacts with pathways relevant in cancer and renal disease. No drugs directly targeting CTDSP2 are approved, but its function and regulation (such as by miR-26a) have been linked to disease mechanisms in solid tumors and metabolic disorders[1][2][3].
